Learning Objectives

During the course of Analytical Chemistry the student will learn some of the most advanced analytical techniques. The most important instrumental methods will be covered as well, in particular those for modern and advanced biological applications. Technical details and applications will be also considered. Particular emphasis will be given to chromatography and mass spectometry.

Program

Units of measurement. Preparation of solutions and dilutions. Validation of analytical methods: The error of measurement: Significant figures. Errors and their detection and minimization. Precision and accuracy. Calibration. Pearson coefficient. Method of least squares. Standardization procedures. Introduction to the sample preparation. Chromatographic methods: distribution coefficient. Column chromatography. The chromatogram and the chromatographic parameters. Chromatographic resolution and efficiency. Band broadening. Van Deemter equation. Basics of gas chromatography. High-performance liquid chromatography: stationary phases and mobile phases. Types of columns. UHPLC and other high-efficiency techniques. Separation mechanisms: absorption, distribution, ion exchange, size exclusion. Ion chromatography. HPLC injector. HPLC detectors: RI, UV-Vis, fluorescence, electrochemical detectors.Mass Spectrometry: ionization techniques: electronic ionization, chemical ionization, electrospray ionization (ESI), matrix effects, multicharged ions, atmospheric pressure chemical ionization (APCI), Matrix-assisted Laser Desorption Ionization (MALDI). General features of a mass spectrum. Mass spectrometric resolution. Fragmentation characteristics. Isotopic cluster. Mass analyzer: quadrupole (Q), ion trap (QIT), time of flight (TOF). Combination of multiple analyzers, QQQ, Q-TOF, TOF / TOF, etc.. Tandem Mass Spectrometry (MS / MS) and collision ion dissociation (CID). Chromatography-mass spectrometry coupling, GC-MS, and HPLC-MS. instrument configurations and operating modes. Matrix effects and their impact on complex samples. HPLC-MS interfaces. Using the mass spectrometer in the analytic field: exact mass, elemental analysis, SIM, SRM, MRM, Scan.

Learning Achievements (Dublin Descriptors)

Knowledge and understanding skills. The student will have to know the main analytical techniques available for the qualitative and quantitative analysis. He/she should also have to know which are the most important substance classes in biological samples. The student should learn the techniques available for sampling, conservation and qualitative and quantitative analysis of chemical components and how to apply them to the real samples. The student should be able to determine what instrumentation is best suited to the needs of the sample. These skills will be verified through written questions.

Ability to apply knowledge and understanding. The student should be able to correctly use the terminology related to analytical chemistry. He will need to master the concentration units. He will need to know how to choose between sampling techniques, sample handling, and analysis to be applied to a biological sample. These skills will be verified through written questions.

Judgment autonomy. The student must be able to make the right decisions on how to deal with an issue related to the analysis of a biological sample.

Communicative Skills. The student should be able to clearly describe the analytical techniques studied with appropriate terms and appropriate examples.

Assessment

Written test with a total of six exercises and open questions on several topics taken from the course program. A 45 minutes time is allowed to complete the test using a form with the six questions and enough space for the answers. The evaluation takes into account the sum of all answers and their overall quality with particular emphasis on correct use of the scientific terminology.
