Learning Objectives

The transmission of valuable information to allow the student to independently deal with the cytometric approach to the diagnosis of the most frequent hematological and non-hematological diseases that can be routinely encountered in the Clinical Pathology laboratory

Program

Review of the structure and functioning of flow cytometers.

Review of the production of monoclonal antibodies and their main uses in laboratory diagnostics.

Relationships between the cytometry laboratory and the hematology laboratory, with notes on the blood counts

Recalls on the CD system.

Role of the main CD antigens in the identification of cell populations of the lymphohematopoietic system.

Basics of cytometric data analysis

Theoretical development of the following points with  computer-guided interpretation of related cases:

1) "Lymphocyte typing"; execution, indications, and meaning

2) Reactive lymphocytosis and neoplastic lymphocytosis

3) Notes on the classification and diagnosis of acute leukemias

4) Role of the cytometry laboratory in autologous bone marrow transplantation

5) Basophil degranulation test (BAT), and its place in allergic diagnostics

6) Cytometric analysis of bronchoalveolar lavage fluid (BALF)

7) Cytometric diagnosis of paroxysmal nocturnal hemoglobinuria

8) Evaluation of DNA in normal and pathological samples. Concepts of cytogenetic ploidy and cytometric ploidy.

Learning Achievements (Dublin Descriptors)

Knowledge and understanding. The student should demonstrate knowledge of the topics listed in the program and be able to understand the relationships between the various information related to them.

Applied knowledge and understanding. The student should prove to be able to interpret the data in his/her possession to understand the diagnostic problem.

Autonomy of judgment. The student should prove to be able to process the data to get a reasoned solution to the diagnostic problem autonomously

Communication skills. The student should prove to be able to communicate clearly and consequentially the diagnostic process followed and the relevance of the final result.

Ability to learn. The student should prove to be able to coordinate the information progressively received during the course, using them logically in a global approach.

Course books

The Course Book consists of the material provided by the teacher and is accessible in Moodle. This material consists of the book "Note di Citometria Diagnostica" and the slides projected in class.

As a reference material, and only for the topics discussed in class, we propose the text "Diagnostica Citometrica delle Neoplasie Ematologiche," Ortolani, Editrice Quattroventi, also uploaded in Moodle.

Still, only for topics discussed in class, another consultation text is "Cellular Diagnostics: Basic Principles, Methods and Clinical Applications of Flow Cytometry," Sack, Tarnok, Rothe eds, Karger 2009. If there are problems with the text availability, don't hesitate to contact the teacher.

Assessment

The final examination consists of the computer analysis of a typical case. The evaluation of the candidate will be expressed as suitable or not suitable.
