Learning Objectives

The course aims to stimulate in the student an attitude of attention towards the chemical phenomenon and to provide the minimum knowledge necessary to operate safely in a laboratory. In fact, in the first part of the course some topics of interest in laboratory practice will be developed, which usually cannot receive due attention in the other theoretical chemistry classes of the degree course. Space was given to the use of equipment and the main laboratory operations, such as classical and instrumental analytical techniques. Through a series of practical exercises, students will be taught to conduct simple experiments with the most common techniques used in a chemical laboratory and to analyze and interpret the results. Each student will be given the protocol of the exercise. The discussion of the experiment will be reported in the laboratory notebook, which will be corrected by the teacher at the end of each exercise to evaluate correct learning. Frontal and laboratory lessons are provided.

Program

The course is divided into theoretical lessons and practical experiences.

1. Safety regulations in the chemistry laboratory:

a) General rules.

2. Common laboratory instruments:

a) Technical and analytical balance; b) glassware; c) vortex; stirring and heating plates; centrifuges, etc.

3. Measurement concept:

a) Concepts of precision and accuracy; b) standard deviation; c) qualitative and quantitative analysis; d) validation of the measure; e) targeted and non-targeted analysis.

4. Weight and volume measurements:

a) Use of scales; b) volume measurements using glass and automatic pipettes.

5. The solutions:

a) Concept of solution; b) concentration of the solutions; c) concentrated solution and dilution procedures; d) preparation of some solutions.

6. Calibration procedures.

a) External standard; b) internal standard; c) standard additions;

7. Sample preparation:

a) Sampling theory and practice. Nature of complex samples; b) Preparation of the sample: theoretical aspects and practical problems in the treatment of real samples and their conservation; c) liquid phase extraction techniques; d) solid phase extraction techniques; e) membrane-based extraction techniques; f) rapid and miniaturized techniques (MEPS, DLLME); g) innovative sample preparation methods: Solid Phase Micro Extraction, QuEChERS, Stir Bar Sorptive Extraction, passive samplers.

8. Introduction to spectroscopic techniques:

a) electromagnetic waves; b) nature of light; c) absorption and emission; d) fluorescence and phosphorescence; e) principles of spectrophotometry.

9. Principles of chromatography:

a) Introduction to chromatography; b) column chromatography; c) chromatogram and chromatographic parameters; d) high performance liquid chromatography (HPLC).

Bridging Courses

It is advisable to take the General Chemistry exam.

In relation to the discipline, the student must show:

D1 Knowledge and understanding

The student will have to demonstrate the possession of a solid knowledge of the fundamentals of general chemistry related to the concept of mole, concentration, unit of measurement of concentrations, solutions, pH and solubility. You will have to show an understanding of the concepts and theory of basic chemistry with particular reference to equilibria in solution.

D2 Ability to apply knowledge and understanding

The student must demonstrate the ability to use, theoretically and practically, the knowledge acquired during the general chemistry course and during the lectures of the Laboratory 1 course (for a total of 28 hours) in all laboratory activities, showing the ability to make connections between the different topics and apply this knowledge to process the data and solve the questions.These skills will be tested during the course with the drafting of the reports relating to each exercise, to be included in the laboratory notebook.

During the laboratory activities (for a total of 28 hours per shift) the teacher will be supported by the laboratory tutor and by a teacher with a supplementary contract.

D3 Autonomy of judgment

The student will have to demonstrate that he is able to critically examine and argue the information contained in the protocol in order to correctly complete the laboratory exercise All this will allow him to develop the aptitude for problem solving that will be verified, both through the drafting of the report , relating to the exercise, in the laboratory notebook, which through targeted application-type questions in the oral exam.

D4 Communication skills

The student must demonstrate the ability to communicate the knowledge acquired to their colleagues and teachers using the specific language of the discipline. These skills will be tested during the oral exam.

D5 Learning skills

The student must show the possession of the learning ability, useful for the continuous deepening of the knowledge of basic chemistry, correctly using the study material provided by the teacher, the recommended textbooks and the in-depth material that can be obtained independently.
